Dysregulation of C-X-C motif ligand 10 during aging and association with cognitive performance

International audience; Chronic low-grade inflammation during aging (inflammaging) is associated with cognitive decline and neurodegeneration; however, the mechanisms underlying inflammaging are unclear. We studied a population (n = 361) of healthy young and old adults from the MyoAge cohort. Peripheral levels of C-X-C motif chemokine ligand 10 (CXCL10) was found to be higher in older adults, compared with young, and negatively associated with working memory performance. This coincided with an age-related reduction in blood DNA methylation at specific CpGs within the CXCL10 gene promoter. The production of colony stimulating factors (CSF) for granulocytes and monocytes is integrated into a network of communicating soluble messenger molecules resulting from T-cell/monocyte interactions. We assessed the capactiy of gamma-Interferon to modulate monocyte secretion of CSF by colony assays and Northern blot analysis to hybridize monocyte RNA with cDNA probes of different CSF-types. Whereas mRNA for GM-CSF was undetectable in untreated and gamma-IFN treated peripheral blood monocytes, the constitutive expression of mRNA for G-CSF and subsequent production of a CSF with biological activities similar to G-CSF could highly be enhanced by exposure of monocytes to gamma-IFN.

ABSTRACTThe performance of the QuantiFERON-cytomegalovirus (CMV) assay was compared to that of a flow cytometry intracellular cytokine staining (ICS) method for the detection of CMV-specific gamma interferon (IFN-γ)-producing CD8+T-cell responses in allogeneic stem cell transplant (allo-SCT) recipients and for estimations of their magnitude and functionality. A total of 90 whole-blood specimens from 23 allo-SCT recipients was analyzed by both methods. Background & Aims Foxp3-expressing regulatory T cells (Tregs) play a key role in the maintenance of the gut immune homeostasis, and an intact transforming growth factor (TGF)-β signaling is required for their function. In inflammatory bowel disease (IBD), the TGF-β signaling is impaired because of high expression of the inhibitory molecule Smad7. Although no intrinsic defects in Tregs function have been shown in IBD, it is still unknown whether colitogenic T cells are susceptible to Treg-mediated suppression.
